From my printed key commands, padlock toggle is "tilde". V is used to revert Player to target view ( becomes target to Player ) You have to be first in Player to target view then press V to revert. You should download the Key Commands pdf document from the OFF website.

Before using the 'Padlock' key, you must Target the next enemy aircraft.
Use the TAC to see which enemy aircraft is highlighted by your Target command and keep punching it to cycle through them. The currently targeted one will be highlighted yellow on the TAC. Then, when you toggle the Padlock view key, your view will be locked on that specific one. Hit Padlock again to toggle it off and go back to regular view. At that point you can just use the padlock key to switch between looking forward and looking at your highlighted target.
Just remember that the Padlock key is an on/off switch so if it's on when you switch the active target, your padlock view will correspondingly snap to the newly selected target and be padlocked on it. That may confuse your orientation if you cycle targets with padlock on. I used to toggle padlock off, then switch targets and turn it back on. Was easier to keep my oriented, along with the selection indicators in the TAC.

...more often than not there's no response when I hit it...
Welcome to "Wings Over Flanders Fields" Forum, Thomas! Perhaps you hit something there - only recently, when I had made a dozen screenshots per F12 key (that's where I put it - maybe it was somewhere else), then it suddenly stopped working. I had pressed F12 three or four times more, but these last screenshots were never made. So, in mid-flight, the key stopped working. Next sortie, it worked fine again.

Olham: TrackIR default profiles typically use the F12 key for centering, so maybe there could be a conflict there per Polovski's previous comment.

No - I have the "Center" command on my joystick's thumb button.

No - I have the "Center" command on my joystick's thumb button. Me too! It seems the best button to use as it is used a lot. I am figuring out which button of 12 to use for the padlock tilde which is awkward as it requires shift.

Hey guys,
thanks to everyone! I tried the Tilde button, but it's awkward on a German keyboard layout (and it didn't work either, as far as I recall, but I don't remember whether I had selected a target then), so I'm relocating it somewhere else, maybe even one of my joystick buttons.
I'll try it out tonight and tell you whether it worked.

I have the button that Cycles the selected enemy air target remapped to a button on my joystick, along with the Padlock toggle. Those two are all you need (along with having the TAC open) to quickly switch targets and toggle the padlock on/off for them.
I recommend putting them on your joystick, if you're going to use them. Because you'll be in a swirling dogfight when you use them.
I've been getting away from using the Padlock since I have TrackIR, but it's an absolute must to have on your joystick if you don't have TIR and can still help even if you do.
